Checklist
Make sure that all the modules Pressure
sensor, Pump, UV and Conductivity are
calibrated.
Set the conductivity reference temperature to
20°C.
No column should be present in the flow path.
Immerse Buffer port A inlet in Buffer A (DM
water).
Immerse Buffer port B inlet in Buffer B (Acetone/
NaCl).
Immerse Sample valve inlet in sample (Acetone/
NaCl).
Fit a 1 ml Sample loop and fill with sample
(Acetone/NaCl).
Connect 2 m of 0.5 mm i.d. tubing to the Waste
position on the Outlet valve.
When performing System performance without
Fraction collector, insert the outlet valve
fractionation tubing into a pre-weighed beaker.
When performing System performance with
Fraction collector, connect the outlet valve
fractionation tubing to Fraction collector with at
least 5 pre-weighed tubes.
Make sure that the system is prefilled with DM
water.
Note: all the required observations by recording
parameters while the performance run is in
progress. Enter the observed values in the ÄKTA
start report template provided in ÄKTA start
