5. Start a photobleach operation by setting the XY Scan button to "time" then begin an
XYT operation;
6. The bleaching is stopped either by manually pressing the 'Scan Stop' button, or by reaching the stop
points defined by the 'Auto Stop' setting or by the number of frames in the Timecourse window.
Pre-bleach

Timecourse: Freerun (no time interval between frames) for 12 frames.
Stimulus: Circular ROI, 4 µs dwell time, 405 nm laser line at 41%.
Activate in series set to 4 pre-bleach frames, 4 bleach frames, leaving 4 frames to be collected post-
bleach.

8.3.2 Avoid spontaneous photoactivation.

A small percentage of photoactivatible fluorescent proteins in the sample will photoactivate when
switching from epi-fluorescence to confocal imaging. Rotating the mirrors (filter cubes) to allow
confocal scanning exposes the entire field of view to a brief flash of UV from the DAPI cube.
Close the manual shutter below the objective turret before switching from epi-fluorescence to confocal
imaging.
8.4

Controlling Computer Display Brightness.

It is often important for one's eyes to become dark-adapted in order to view the specimen through the
eyepieces, or even on the computer display. Glare from the computer desktop and software windows
can be a nuisance. The desktop display on the confocal computer has been set to black. The Fluoview
software application has an option called "Darkroom" that will set the white background of all menus
and windows to a dark gray. It does take getting used to the appearance.
