7.2.2 DIC polarizer.

This is on the condenser, Figure 13.4. This must be slid into the halogen path in order to obtain a DIC
image. Pull it out to obtain a plain brightfield image. The position of the polarizer has no effect on
confocal fluorescent imaging.

7.2.3 DIC mirror.

This component is controlled automatically by the Microscope Controller. Selecting the DICT button
also rotates a wheel in the condenser that rotates the appropriate Wallaston prism into place.
7.3

Setting Koehler Illumination.

Maximal resolution and field uniformity with brightfield and DIC is only achieved if the condenser is in
focus to the same plane as the objective lens. Also, a focused condenser acts as the condenser lens for
the transmitted light detector. A properly configured condenser will make brightfield or DIC much more
